Abstract:

Introduction: Previous studies have confirmed the sidelong effects of Doxorubicin (DOX) on healthy tissues, but pretreatment effect of regular Aerobic exercise in restrain of hepatotoxicity induced by doxorubicin is not clear. The aim of this study was to investigate the pretreatment effect of regular aerobic exercise on hepatic oxidative stress induced with doxorubicin of Wistar rats in different age groups. Methods: In the experimental study, 72 male wistar rats divided into three groups of age (three and 30 months) were again randomly separated into subgroups: control+DOX (20 mg/kg, n=8), and Training+DOX (T+DOX20mg/kg, n=8) Training+ Saline (T+S) groups. The training protocol included treadmill running progressively between 25 to 39 min/day and 15 to 17m/min, 5 days/week for 3 weeks. DOX and saline injection was performed 24 h after the last exercise session, and tissue collection was done 24 h after the injections. Results: The results showed that the DOX-induced did not significant effect on HSP70 of liver tissue of rats in different ages (young, middle-aged and elderly) (P=0.272). DOX-induced had a significant effect on MDA (P=0.002), PC (P=0.002) and GPx (P=0.001) of liver tissue of rats in different ages (young, middle-aged and elderly). Pre-treatment with aerobic exercise had a significant effect on changes of HSP70 (P=0.001), MDA (P=0.001), PC (P=0.001) and GPx (P=0.001) in DOX-induced liver in different ages rats (young, middle-aged and elderly. There was any significant difference between the levels of HSP70 (P=0.741), MDA (P=0.539), PC (P=0.075) and GPx (P=0.071) liver tissue in different ages (young, middle-aged and elderly) with pre-treatment and aerobic exercise. Conclusion: Our study suggests that liver protection induced by chronically exercise in DOX treated rats is associated with inhibition of oxidative stress and increase in the efficiency of antioxidant system.

Age-Related Changes in Doxorubicin-Induced Oxidative Damage: Protective and Pretreatment Effects of Short-Term Aerobic Exercise

ABSTRACT        Background and Objective: Doxorubicin (DOX) is an effective anticancer drug. It has been shown that  a short-term exercise performed prior to DOX-treatment has no effect on cardiotoxicity in young rats. In the present study, old and young rats were evaluated to determine the protective effects of pre-treatment with short-term exercise on D...

Is sub-chronic exercise in Combination with medicinal nanoparticles a protective strategy against Doxorubicin-induced Hepatic oxidative stress and apoptosis in aging model rats?

Objective(s): Oxidative stress and apoptosis are the major side effects of doxorubicin (DOX) and the advantages accruing fromexercise and some medicinal herbs in mitigation of these toxic side effects is well documented. But so far, the effects of exercise in combination with medicinal nanoparticles on oxidative stress and apoptosis signaling simultaneously, in liver tissue are unknown. Hence, ...

Protective Effects of Aerobic Training and Crocin on Doxorubicin Induced Heart Tissue Oxidative Stress in Male Rats

Background and Objectives: Doxorubicin-increased oxidative stress destroys tumors and normal cells, especially in heart tissues. The objective of the present study was to investigate effects of eight weeks of aerobic training and crocin consumption on doxorubicin induced heart tissue oxidative stress in male rats.  Materials & Methods: In this experimental study, 50 male rats were randomly div...

Age Related Changes in NAD+ Metabolism Oxidative Stress and Sirt1 Activity in Wistar Rats

The cofactor nicotinamide adenine dinucleotide (NAD+) has emerged as a key regulator of metabolism, stress resistance and longevity. Apart from its role as an important redox carrier, NAD+ also serves as the sole substrate for NAD-dependent enzymes, including poly(ADP-ribose) polymerase (PARP), an important DNA nick sensor, and NAD-dependent histone deacetylases, Sirtuins which play an importan...

Moderate aerobic exercise training decreases middle-aged induced pathologic cardiac hypertrophy by improving Klotho expression, MAPK signaling pathway and oxidative stress status in Wistar rats

Objective(s): This study aimed to investigate the effect of aerobic training on serum levels of Klotho, cardiac tissue levels of H2O2 and phosphorylation of ERK1/2 and P38 as well as left ventricular internal diameter (LVID), the left ventricle wall thickness (LVWT) and fibrosis in middle-aged rats. Materials and Methods: Forty wistar rats, including young rats (n=10, 4 month-old) and middle-ag...

Amitriptyline Effects on Pentylentetrazole-Induced Oxidative Stress in Epileptic Male Wistar Rats

Background: Drug-induced epilepsy can be caused by certain medications and chemical agents. Oxidizing agents are among the chemical compounds that cause epilepsy. The aim of this study was to evaluate the anticonvulsant and antioxidant effects of amitriptyline on pentylenetetrazole (PTZ)-induced seizure in male rats. Materials and Methods: In this experimental study, 28 male Wistar rats (20...
